Mechanism of Action:
Inside cells, fludarabine is transformed into 2-fluoro-ara-ATP, its active metabolite, which is essential to its anti-cancer properties. This metabolite prevents the synthesis of new DNA by inhibiting the enzymes ribonucleotide reductase and DNA polymerase, which are necessary for DNA synthesis. Furthermore, it results in chain termination after DNA replication, which eventually causes rapidly dividing cancer cells to undergo apoptosis. Fludarabine works well to treat hematological cancers such as chronic lymphocytic leukemia (CLL) and some forms of non-Hodgkin's lymphoma because of this mechanism.
